چکیده
Abstract H5N1 vaccines adjuvanted in the oil-in-water emulsion MF59 are maintained Strategic National Stockpile for pre-pandemic preparedness. The US population currently naïve to H5 and a prime/boost regimen will likely be needed engender protective response. Ideally, rapid response pandemic influenza would benefit from being able induce protection single dose of vaccine. Here, we have used systems immunology approach mice examine effect formulating recombinant AddaVax alone or with CpG+MPLA. does not stimulate innate immune system, although oil droplets do act as nanocarrier antigen enhance B cell activation, presumably by BCR cross-linking. In vitro, germinal center reactions show inclusion CpG+MPLA (a combination adjuvant call IVAX-1) synergize maturation class-switching. Similarly, vivo, H5/IVAX-1 enhances both H5-specific T responses compared individual components. Single mRNA sequencing spleen cells after boost indicate H5/CpG+MPLA more similar each other but distinct H5/AddaVax H5/PBS. Unexpectedly, H5/AddaVax, all engendered 100% efficacy against lethal intranasal challenge virus, only dose. Our data potential certain adjuvants immunogenicity achieve vaccination. Matrix‐M adjuvanted virosomal H5N1 vaccine confers protection against lethal viral challenge in a murine model
BACKGROUND A candidate pandemic influenza H5N1 vaccine should provide rapid and long-lasting immunity against antigenically drifted viruses. As H5N1 viruses are poorly immunogenic, this may require a combination of immune potentiating strategies. An attractive approach is combining the intrinsic immunogenicity of virosomes with another promising adjuvant to further boost the immune response. As...
متن کاملCross-Protection against Lethal H5N1 Challenge in Ferrets with an Adjuvanted Pandemic Influenza Vaccine
BACKGROUND Unprecedented spread between birds and mammals of highly pathogenic avian influenza viruses (HPAI) of the H5N1 subtype has resulted in hundreds of human infections with a high fatality rate. This has highlighted the urgent need for the development of H5N1 vaccines that can be produced rapidly and in sufficient quantities. Potential pandemic inactivated vaccines will ideally induce su...
متن کاملVaccination with different M2e epitope densities confers partial protection against H5N1 influenza A virus challenge in chickens.
OBJECTIVE Currently, research is focused on universal influenza vaccines based on various ectodomains of the influenza matrix protein 2 (M2e). Such vaccines are tested mostly using mouse-adapted influenza viruses and in mouse or ferret models. The aim of this study was to investigate in a chicken model the protective efficacy of vaccines based on avian-type M2e at different epitope densities. ...
متن کاملTransient Humoral Protection against H5N1 Challenge after Seasonal Influenza Vaccination of Humans
Current influenza vaccines are believed to confer protection against a narrow range of virus strains. The identification of broadly influenza neutralizing antibodies (bnAbs) has triggered efforts to develop vaccines providing 'universal' protection against influenza. Several bnAbs were isolated from humans recently vaccinated with conventional influenza vaccines, suggesting that such vaccines c...
متن کاملEmulsified Nanoparticles Containing Inactivated Influenza Virus and CpG Oligodeoxynucleotides Critically Influences the Host Immune Responses in Mice
BACKGROUND Antigen sparing and cross-protective immunity are regarded as crucial in pandemic influenza vaccine development. Both targets can be achieved by adjuvantation strategy to elicit a robust and broadened immune response. We assessed the immunogenicity of an inactivated H5N1 whole-virion vaccine (A/Vietnam/1194/2004 NIBRG-14, clade 1) formulated with emulsified nanoparticles and investig...
